Hi,
There are different ways to back up Active Directory using Microsoft tools which was previously known as NTbackup or using other third party backup tool.
You can automate your backup through Windows Server Backup using Task Scheduler. If you are using any other backup tool so it will automatically take systemstate backup while taking server (domain controller) backup.
You can only perform a system state backup using the WBADMIN.EXE command-line program—the MMC snap-in doesn't provide this option. To perform a system state backup, you can use this command:
C:\> wbadmin start systemstatebackup –backuptarget:e:
Note:
Before you can backup Server 2008 you need to install the backup features from the Server Manager.
- To install the backup features click Start→ Server Manager.
- Next click Features→ Add Features
- Scroll to the bottom and select both the Windows Server Backup and the Command Line Tools
- Click Next, then click Install

It will not delete any data or format drive. This command will only keep the backup on E drive.
